摘 要:目的:通过对栀子炮制前后含量变化较大的两种色素类成分藏红花酸糖苷-1(crocin-1)和藏红花酸(crocetin)进行药效学比较,探讨药理作用变化的物质基础。方法:采用炎症,凝血,化学性肝损伤和血液流变学等方法,观察两种成分的作用差别。结果:两种成分对小鼠炎症和四氯化碳造成的肝损伤无明显作用;藏红花酸糖苷-1中,大剂量和藏红花酸各剂量能明显降低血瘀症大鼠高切变率下的血液黏度、缩短凝血酶原时间(PT);藏红花酸中、大剂量同时缩短活化部分凝血活酶时间(APTT)。结论:藏红花酸糖苷-1、藏红花酸抗炎、保肝作用不明显,对高切变率下的全血黏度有明显降低作用,缩短PT。藏红花酸大剂量凝血作用强于藏红花酸糖苷-1。提示藏红花酸与栀子炒焦后增强止血作用有关。Objective: To explore the pharmacodynamical active substances of Gardenia jasminds ELLis, the comparative pharmacodynamic study was carried out on crocin-1 and crocetin. Methods: To observe the different pharmacological influence of crocin-1 and crocetin on anti-inflammatory, blood coagulation, hepatic injury and hemorheological changes. Results: Neither crocin-1 nor crocetin has shown obvious effects of anti-inflammaory or protective effect against hepatic injury induced by CCl4 in mice. Crocin-1 and crocetin can decrease the blood viscosity under high shear stress, and curtail prothrombin time(PF). Both medium and high concentration groups of crocetin can curtail the thrombin time and activated partial thromboplastin time(APTT). Conclusions: Neither Crocin-1 nor crocetin has shown obvious effects on anti-inflammatory,hepatic protection. Both components Can reduce the whole blood viscosity and PT significantly. Crocetin of high concentration showed strong effects on promoting coagulation than crocin-1. The results suggest that erocetin may be the substance matter of hemostasis action of fructus gardeniae.
