摘 要:以琼脂糖和尼龙66微孔滤膜为原料,发展了两种用于去除内毒素的不同类型的亲和介质。在对原料进行化学改性后,键合了含六个碳原子的间隔臂和组氨酸亲和配基。它们对内毒素的亲和容量分别为 2.0 mg/g和 0.2 mg/g,内毒素和介质之间的表观解离常数分别为8.1×10-10mol/L和3.0×10-8mol/L。在所选择的最佳作用条件下,可成功地应用于去除医用水,氨基酸注射液,牛血清白蛋白,溶菌酶等生物医药制剂中的内毒素。Two different kinds of affinity matrixes consisting of agarose and nylon 66 micro- filtration membrane for endotoxin removal were developed. After crosslinking of sepharose. 4B with epichlorohydrin, a space arm with 6 carbon atoms was bound on it. After that, the column affinity material was coupled with histidine ligand by the glutaraldehyde method. Based on Langmuir adsorption curve of this matrix obtained by batch test method, its affini- ty capacity for endotoxin was 2. 0 mg per gram. The apparent dissociation constant between endotoxin and the adsorbent was 8. 1 ×10-10 mol/L. With a similar method, a ny1on 66 mi- crofiltration affinity membrane was prepared and its characteristics were investigated. The adsorption capacity of this membrane was 0. 2 mg of endotoxin per gram membrane. Its ap- parent dissociation constant was 3. 0×10-8 mol/L. The column and membrane separator Pre- pared by these two affinity matrixes could be used to remove endotoxin in medical water and some biomedical products, such as injection so1ution of amino acids, bovine serum albumin and lysozyme at selected optimum conditions.
