摘  要:目的探索巯基乙酸(TGA)对非洲蟾蜍和小鼠卵母细胞体外成熟的影响。方法用0μg/ml、5μg/ml、25μg/ml和125μg/mlTGA处理非洲蟾蜍卵母细胞,体外培养,并观察生发泡破裂(GVBD)发生情况。用0μg/ml、5μg/ml和50μg/ml的TGA处理小鼠卵母细胞,体外培养,观察GVBD和第一极体(PB1)释放的情况。结果在非洲蟾蜍卵母细胞中,TGA处理2h、4h和6h后,各TGA处理组的GVBD率均高于对照组,但在8h时,TGA处理组与对照组GVBD率之间无统计学差异。在小鼠卵母细胞中,仅12h时5μg/ml组的GVBD率高于对照组,但在各时间点,TGA处理组的GVBD率都有增加趋势;在16h,5μg/ml和50μg/mlTGA处理可促进PB1释放,但与对照组相比,仅5μg/ml组有统计学意义。结论结果表明,TGA可促进非洲蟾蜍卵母细胞第1次减数分裂的启动。在小鼠中卵母细胞中,低剂量的TGA也可加速第1次减数分裂的启动。低剂量的TGA可增加小鼠卵母细胞的成熟率;但高剂量的TGA却没有明显的促进作用,推测TGA对小鼠卵母细胞成熟的影响可能存在双重性。Objective To investigate the effect of thioglycolic acid on oocyte maturation process in vitro in mice and Xenopus. Methods Xenopus oocytes were treated with TGA in vitro at dose of 0 μg/ml, 5 μg/ml, 25 μg/ml and 125 μg/ml, while mice oocytes at 0 μg/ml, 5 μg/ml and 50 μg/ml. GVBD of Xenopus and mice oocytes, and PB1 exclusion of mouse oocytes were observed during culture process. Results In Xenopus oocytes, GVBD rate in TGA-treated groups was higher compared with control group, at 2 h, 4 h, and 6 h after TGA treatment, while there was no difference between TGA-treated and control group at 8 h. In mouse oocytes, GVBD rate in TGA-treated groups was higher than control group, although despite that difference existed only in 5 μg/ml TGA group at 12 h. At 16 h, 5 μg/ml and 50 μg/ml TGA could elevate PB1 exclusion rate compared with control group, with 5 μg/ml having statistical difference. Conclusions These results indicate that TGA treatment can accelerate GVBD in Xenopus oocytes, and GVBD of mouse oocytes at low dose. Low TGA treatment can also promote mouse oocyte maturation, but high dose has no significant maturationpromoting role, suggesting that TGA may have a dual-role on mouse oocyte maturation through different pathways.
