摘 要:目的通过线粒体孵育液染锰并用NAC干预研究锰对大鼠线粒体能量代谢的影响及NAC的防护效果。方法Wistar大鼠12只,麻醉后断头取脑,梯度离心得线粒体液。将所得线粒体液分为6组,第1组为对照组;第2。5组为不同剂量染锰组,分别加入5、50、500、1000μmol/LMnCl2;第6组为NAC预处理组,线粒体液先用500μmol/LNAC预处理1h后加500μmol/LMnCl2。各组于培养箱内30℃孵育2h后测顺乌头酸酶、线粒体复合体Ⅰ的活性和线粒体膜电位,每个指标重复测量6次。结果与对照组相比,500、1000μmol/L染锰组顺乌头酸酶活性显著降低(P〈0.05),50、500、1000μmol/L染锰组线粒体复合体Ⅰ的活性和线粒体膜电位显著降低(P〈0.05),且在一定范围内随着MnCl2剂量增加,顺乌头酸酶、线粒体复合体Ⅰ的活性和线粒体膜电位有降低趋势。与单纯染锰组相比,NAC预处理组顺乌头酸酶、线粒体复合体Ⅰ的活性和线粒体膜电位显著升高(P〈0.05)。结论锰可以剂量依赖性地诱导大鼠脑线粒体能量代谢降低;NAC预处理能预防锰所致大鼠脑线粒体能量代谢降低。Objective To elucidate the effect of MnCl2 exposure on energy metabolism of brain mitochondria of rat and the preventive effect of NAC in vitro. Method Mitochondria from Wistar rats' whole brain were prepared using gradient eentrifugation, then were incubated with different concentrations of MnCl2 (5 μmol/L, 50 μmol/L, 500 μmol/L, 1 000 μmol/L) at 30℃ for 2 h; only one group of mitochondria was pretreated with 500 μmol/L of NAC for 1 h, and incubated with 500 μmol/L of MnCl2 again. The activities of aeonitase, mitochondrial complex I and the membrane potential of mitochondria were detected. Result The results showed that the aeonitase activities in 500 μtool/L, 1 000 μmol/L MnCl2 groups were reduced, and the mitochondrial complex I activity, mitochondrion membrane potential in 50, 500, 1 000 μmol/L MnCl2 groups were also significantly decreased compared to the controls ( P 〈 0. 05 ). While in 500 μmol/L NAC pretreated group, the activities of aeonitase, mitochondrial complex I and the membrane potential of mitochondria were all increased compared to 500 μmol/L MnCl2 group ( P 〈 0. 05 ). Conclusion It is suggested that manganese could dose-dependently induce the decline of energy metabolism of brain mitochondria in rat; this change could be prevented by pretreating with NAC.
