摘 要:[目的]了解菲律宾菌素减少人类疱疹病毒-6型包膜胆固醇后,对病毒感染性的影响。[方法]用不同浓度的菲律宾菌素作用于HHV-6A GS株,经20%蔗糖缓冲液梯度离心,去除菲律宾菌素并纯化病毒,分别感染HSB-2和Jurkat细胞,然后采用IFA法、Hoechst 33258核染色及免疫蛋白杂交等方法检测病毒受菲律宾菌素作用后,其对靶细胞的融合、进入的影响。[结果]菲律宾菌素能减少HHV-6A包膜胆固醇,病毒的感染性及诱导细胞融合的能力显著减弱。[结论]HHV-6A包膜胆固醇在细胞融合过程中起着重要作用,同时也是病毒进入宿主细胞的关键因素。[Objective]To find out effects on Human Herpesvirus-6(HHV-6) infectivity after Filipin Ⅲ decreasing viral envelope cholesterol.[Methods]HHV-6A GS strains were treated with different concentrations of Filipin Ⅲ.The treated viruse was subjected to gradient centrifugation through 20% sucrose cushion to remove Filipin Ⅲ and purified virus,and HSB-2 cells and Jurkat cells were infected by treated and untreated HHV-6A respectively.The roles of HHV-6A induced cell-fusion and the ability of entery were detected by IFA,DNA stained with Hoechst 33258 and Western blotting post HHV-6A cholesterol depletion.[Results]Filipin Ⅲ reduced cholesterol in the HHV-6 envelope,and markedly affected HHV-6 infectivity and HHV-6-induced cell fusion.[Conclusion]the cholesterol present in the HHV-6 envelope plays a prominent role in the fusion process and is a key component in viral entry.
