缺氧对膀胱平滑肌细胞缝隙连接蛋白40和缝隙连接蛋白43的影响

摘　　要：目的:探讨缺氧环境下缝隙连接蛋白(Cx40、Cx43)在mRNA水平上的变化,研究细胞缺氧和Cx40、Cx43表达变化在不稳定膀胱发病机理上的相关关系。方法:体外培养大鼠膀胱平滑肌细胞,建立细胞缺氧模型;在细胞缺氧1 h后收集细胞,运用逆转录-聚合酶链反应技术分析Cx40、Cx43。结果:缺氧1 h,Cx40、Cx43在mRNA水平上表达较正常氧对照组有明显升高(P<0.01)。结论:细胞缺氧会引起Cx40、Cx43在mRNA水平上表达的变化,这可能与不稳定膀胱的发生有密切联系。Objective： To observe the connexin（Cx40, Cx43）changes under cell hypoxia in smooth muscle cells. Method： The smooth muscle cells were cultured in vitro. The mRNA levels of Cx40 and Cx43 were detected by RT-PCR one hour after cell hypoxia. Results： The mRNA levels of Cx40 and Cx43 in the one hour cell hypoxia group were higher than those in the cell normoxia group（P 〈 0.01）. Conclusion： Cell hypoxia can cause the increase of mRNA of Cx40 and Cx43, and it might be contributed to the unstable bladder.
