摘 要:目的:研究利多卡因对缺氧/复氧培养大鼠心肌细胞LDH漏出及c-fos表达的影响。方法:建立SD大鼠心肌细胞原代培养及缺氧/复氧模型。将40孔原代培养大鼠心肌细胞分为5组,每组8孔。Ⅰ组为对照组;Ⅱ组缺氧2h复氧3h;Ⅲ、Ⅳ和Ⅴ组与Ⅱ组比在缺氧前开始分别加入利多卡因50μmol.L-1、100μmol.L-1和150μmol.L-1。复氧3 h时用生化比色法测定培养基中乳酸脱氢酶(LDH)活力。将75孔细胞同上分为5组,在复氧2 h时用免疫组化法和图像平均灰度分析法测定c-fos的表达。结果:3种浓度利多卡因均能减少心肌细胞LDH漏出(P<0.05)。Ⅱ组与Ⅰ组比c-fos表达明显增加,Ⅵ组和Ⅴ组与Ⅱ组比较c-fos表达明显被抑制,平均灰度值差异有统计学意义(P<0.05)。结论:3种浓度的利多卡因能防治心肌细胞缺氧/复氧损伤,并且呈剂量相关性。Objective: To observe the effect of lidocaine on c-los expression in cultured cardiac myocytes of neonatal rats and the concentration of lactate dehydrogenase(LDH) in culture medium during reoxygenation. Methods: Original cultured cardiomyocytes of SD rat and hypoxia-reoxygenation injuried immature cariomyocyte were established. The 40 caves of cultured cardiac myocytes were divided into 5 groups: group Ⅰ as control;group Ⅱ as hypoxia-reoxygenation (hypoxia 2 hours-reoxygenation 3 hours)control; With eomparaded to group Ⅱ ,the groups Ⅲ、Ⅳ, Ⅴ and Ⅵ were treated with lidocaine(50 μmol·L^-1, 100 μmol·L^-1 and 150 μmol·L^-1) since hypoxia respectively. Immunohistochemistry and mean gray were used to estimate the effect of lidocaine on expression of c-los in cultured rat cardiomyocyte of each group 2 hours after reoxygenation. Release of lactate dehydrogenase of the cardiomyocytes of each group was investigated and determined as injury index of cardiomyocytes. Results: Release of lactate dehydrogenase was inhibited by lidocaine in 3 kinds of density,and expression of c-los of the cardiomyocytes was significantly inhibited by lidocaine in 100 μmol·L^-1 and 150 μmol ·L^-1 (O〈0.05) .Conclusion: Hypoxia-reoxygenation injury of the cardiomyocyte was released, then the expression of c-fos during reoxygenation was inhibited by lidocaine.
