摘 要:目的探讨新生大鼠嗅鞘细胞的原代培养及纯化方法,并对其形态学特征进行观察。方法分离培养新生大鼠嗅球中的嗅鞘细胞,联合应用差速贴壁法和阿糖胞苷抑制法进行纯化,NGFRp75免疫组化染色鉴定细胞。结果可获得较高纯度的嗅鞘细胞,其形态主要为双极、多极和无突起的"油煎蛋"形,细胞突起细长并交织成网状。结论该方法步骤简便,可重复性好,具有较高的实用价值。Objective To investigate the method of primary culture and purification for olfactory ensheathing cells (OECs) and to observe the morphologic features of OECs. Methods OECs were isolated and cultured from neonatal rat's olfactory bulbs, and purified by the combined use of the different rates of attachment and Ara-c inhibition. OECs were identified by nerve growth factor receptor p75 (NGFRp75) immunohistochemical staining. Results High purity of OECs could be obtained. The cultured OECs presented three morphological types: bipolar, multipolar cells and fried egg-like cells. The processes of OECs were long and thin and connected into a net. Conclusion This method has highly practical value because of its convenient procedure and reproducibility.
