摘 要:目的:研究红景天苷对麻醉开胸犬冠状动脉结扎所致心肌缺血的保护作用。方法:麻醉犬冠状动脉结扎造成心肌缺血模型,随机分为对照组、模型组、溶剂组、地尔硫注射液(0.8 mg/kg)组、葛根素注射液(4.0 mg/kg)组、红景天苷高、中、低(8.0,4.0,2.0 mg/kg)剂量组。采用心外膜标测和定量组织学硝基四氮唑蓝(NBT)染色法测定心肌损伤范围及损伤程度,并取结扎部位下1 cm处心肌组织作病理检测。测定血浆中的丙二醛(MDA)和超氧化物歧化酶(SOD)活性。结果:红景天苷可显著减轻由心外膜电图所标测的心肌缺血程度及心肌缺血范围;可显著减小通过NBT染色所显示的梗死区;可显著减少血浆中MDA的含量,增强SOD的活性。结论:红景天苷对麻醉犬冠脉结扎所致心肌缺血有显著的保护作用。Aim:To study the effects of salidroside on acute myocardial isehemia induced by ligating coronary artery of the anaesthetic thoraco-opening dogs. Methods: The model of the myocardial ischemia of the anaesthetic thoracoopened dogs was developed by ligating the left coronary anterior descending artery. The dogs were randomly divided into eight groups: control, model, solvent, diltiazem hydrochloride for injection (0.8 mg/kg), puerarin for injection (4.0 mg/kg) and salidroside for injection(8.0,4.0, and 2.0 mg/kg). The epicardial electrocardiogram and quantitative histological analysis combined with nitroblue tetrazolium(NBT) staining method were used to determine the range and degree of the damage induced by myocardial ischemia. And the myocardium tissue at the part of 1 cm under the ligated location was taken for the pathological detection. The activities of both plasma malondialdehyde (MDA) and superoxide dismutase (SOD) were determined. Results: Salidroside showed to significantly decrease the severity of myocardial ischemia and reduce the extent of the infarction. It was also found to ameliorate the activities of the plasma MDA, and to enhance the aetivities of the plasma SOD. Conclusion: Salidroside could significantly protect the ischemic myocardium of the anesthetic thoraco-opening dogs induced by ligating coronary artery.
