低温保存肝脏肝功能与肝细胞糖原关系的研究

摘　　要：目的:探讨低温保存肝脏肝功能与肝细胞糖原含量之间的关系。方法:以兔为实验对象,术前24 h禁食、普食及普食加静脉注射葡萄糖等方法制备4组糖原含量不同的供肝模型。于供肝恢复血流2 h时,获取受体动物血液标本,以测定各组动物的肝功能状况。结果:4组供肝切取前组织内糖原含量,A组(15.42±2.60)mg/g,B组(51.83±6.61)mg/g,C组(63.22±5.84)mg/g,D组(78.58±8.46)mg/g,D组高于C组(P<0.05),各组彼此间均有显著差异(P<0.05)。供肝再灌注2 h时,4组受体兔血液中AST、ALT及AKP含量变化均表现为A组>B组>C组>D组,且各组彼此间均有显著差异(P<0.05)。结论:在供肝切取前增加供肝肝细胞内的糖原含量,有望减轻供肝冷缺血损伤程度,提高肝移植的成功率。Objective： To study the function of transplanted liver and content of hepatic glycogen in donor livers stored in low temperature. Methods： 56 rabbits were divided into liver donor group and liver recipient group. The two groups were further divided into four subgroups, i. e. fasted 24 h group（group A）, common feed group（group B）, common feed concurrently with intravenous injection of glucose 4 times group（group C） and 6 times group（group D）. The livers of donor group were taken away and stored in 4 ℃ for 9 h. By the transplanted livers regained blood supply for 2 h, the samples of blood of recipient groups were collected; the liver functions and content of hepatic glycogen of recipient group were examined. Results：The contents of hepatic glycogen in donor liver from group A was （15. 42±2. 60）mg/g, group B was（51.83±6. 61）mg/g, group C was（63. 22±5. 84） mg/g and group D was（78. 58±8. 46）mg/g, respectively, and the differences among four groups were significant（P〈0.05）. The changes of levels of AST, ALT and AKP in blood samples were group A〉group B〉group C〉group D and the differences among four groups were significant （P〈0. 05）. Conclusion： Before donor liver is taken away, addition supply of hepatic glycogen may be helpful to relieve cold ischemic damage of donor liver and elevate the achievement ratio of liver transplantation.
