摘 要:运用PCR技术和相关生物软件,对大头蛙Limnonectes kuhlii和脆皮大头蛙Limnonectes fragilis的线粒体coⅠ基因全序列进行了测定与分析.结合GenBank上已公布的无尾目10个科22个物种的同源序列,以爬行纲楔齿蜥Sphenodon punctatus和密河鳄Alligator mississippiensis为外群,基于核苷酸序列(选取前两位密码子)和氨基酸序列两种数据形式,分别构建NJ树和MP树.结果表明:系统树将无尾目10个科分为两大支,一支为新蛙亚目Neobatrachia的5科15个物种,一支为古蛙亚目Archaeobatrachia的5个科9个物种(其中包括锄足蟾科Pelobatidae和负子蟾科Pipidae),并推测这两个亚目均为单系群.古蛙亚目一支内的盘舌蟾科Discoglossidae与铃蟾科Bombinatoridae亲缘关系较近;新蛙亚目内蟾蜍科Bufonidae与雨蛙科Hylidae,蛙科Ranidae与树蛙科Rhacophoridae两两亲缘关系较近.蛙科中泽陆蛙Fejervarya limnocharis与大头蛙属3个种首先形成姐妹群,表明其较近的亲缘关系,同时推断大头蛙与福建大头蛙Limnonectes fujianensis亲缘关系近于脆皮大头蛙.The complete sequences of mitochondrial coⅠ genes of Limnonectes kuhlii and Limnonectes fragilis were amplified and sequenced using PCR technique, and then analyzed through several biologic softwares. When Sphenodon punctatus (AF534390) and Alligator mississippiensis (Y13113) were selected as outgroups, we aligned them with the same mtDNA fragments of 22 other frogs recorded in the Gen- Bank, and reconstructed phylogenetic trees using MEGA version 3.1, based on the nucleotides sequences (1st+2nd codon positions) and the amino acid sequences data, respectively. Neighbor-joining (NJ) and Maximum Parsimony (MP) analyses generated nearly identical tree topologies. The results were revealed: The species examined in this study were clusterd into two clades. The first clade included 5 families of Ar- chaeobatrachia (9 Species, including Pelobatidae and Pipidae), and the second clade included 5 families of Neobatrachia (15 Species). The results supported the monophyly of each of the two suborders (Archaeobatrachia and Neobatrachia). A close relationship between Discoglossidae and Bombinatoridae was showed within Archaeobatrachia. There were two pairs of taxa cluster: Bufonidae + Hylidae and Ranidae + Rhacophoridae in Neobatrachia. Two trees strongly supported a sister group relationship for 3 species of Limnonectes (L. fujianensis, L. kuhlii and L. fragilis) and Fejervarya (F. limnocharis), and favored a L. fujianensis + L. kuhlii clade to exclusion of L. fragilis.
