降钙素基因相关肽对血管平滑肌细胞增殖及小凹蛋白1表达的影响

摘　　要：目的观察降钙素基因相关肽在抑制血管平滑肌细胞增殖或肥大过程中是否伴有诱导其凋亡的作用,并探讨其作用机制是否涉及到血管平滑肌细胞内小凹蛋白1和caspase-3的变化。方法采用贴块法体外培养大鼠胸主动脉血管平滑肌细胞,增殖或肥大型血管平滑肌细胞通过血管紧张素Ⅱ刺激静止期血管平滑肌细胞24 h建立。用噻唑蓝比色法和流式细胞仪分析血管平滑肌细胞增殖、凋亡及细胞周期变化,吖啶橙/溴化乙啶染色观察细胞及细胞核形态改变,Western blotting法测定不同状态下血管平滑肌细胞内小凹蛋白1和caspase-3的表达。结果降钙素基因相关肽能降低血管紧张素Ⅱ诱导的增殖型血管平滑肌细胞的代谢活性、细胞周期增殖指数(P<0.05);形态学及流式细胞仪分析表明降钙素基因相关肽对增殖型血管平滑肌细胞无明显的诱导凋亡作用。同时发现降钙素基因相关肽使血管平滑肌细胞内小凹蛋白1的表达增加,而对caspase-3的表达无明显影响。结论降钙素基因相关肽能显著抑制血管紧张素Ⅱ诱导的血管平滑肌细胞增殖,但诱导血管平滑肌细胞凋亡的作用较小;其抑制增殖的细胞内信号传导途径可能与增强小凹蛋白1表达有关。Aim To observe the effect of calcitonin gene-related pepfide { CGRP） on the proliferation or apoptosis of hy- perplasia / hypertrophic vascular smooth muscle cells （ HVSMC ） induced by angiotensin Ⅱ （ Ang Ⅱ ）, and explore whether the mechanism is related to the expression of caveolin-1 and caspase-3. Methods The HVSMC was established by quiet rat vascular smooth muscle cells incubated with Ang II for 24 hours. MTT and flow cytometry were used to determine the ability of proliferation of HVSMC induced by Ang II in the presence or absence of CGRP. Western blotting was used to determine the expression of caveolin-1 and caspase-3. Results Treatment with CGRP for 8 h could inhibit the viability, continuous proliferation of HVSMC induced by Ang Ⅱ, but it could not significantly induce the HVSMC apoptosis. Tiffs effect of CGRP was concomitant with the increased expression of cavcolin-1 and with no significant change on the expression of caspase-3. Conchusion CGRP could significantly inhibit the proliferation of HVSMC induced by Ang Ⅱ, but not significantly induce apoptosis of HVSMC, the signal pathway of inhibitory, effect of CGRP may be related to the increase of the cavcolin-1 expression.
