摘  要:目的:探讨过氧化物酶增殖物激活受体PPARγ(Peroxisome proliferator-acti-vated receptor-γ)及其配体对滋养细胞肿瘤细胞系JEG-3浸润能力的影响。方法:通过免疫荧光细胞化学方法检测JEG-3细胞系中PPARγ的表达,采用Transwell侵入系统检测PPARγ的天然激动配体15脱氧-前列腺素J2(15d-PGJ2)对滋养细胞浸润能力的影响。结果:在JEG-3滋养细胞系中有PPARγ蛋白表达,PPARγ激动配体15d-PGJ2处理后浸润穿膜细胞数明显减少,与对照比较差异有统计学意义(P<0.01),且具有剂量依赖关系。结论:PPARγ被配体15d-PGJ2激活后抑制JEG-3滋养细胞浸润能力,为今后PPARγ配体在滋养细胞浸润异常有关的妊娠并发症治疗中可能的临床应用提供理论和实验基础。Objective:To investigate PPARγand its ligand's contribution to the infihrating ability of JEG-3 cell line. Methods: The expression of PPARγin JEG-3 cell line was detected by immunofluorescence cytochernistry method. Tanswell Invasion system was used to detect the influence of PPARγ's natural emotional ligand 15-d-PGJ2 to the infiltrating ability of the nutriant cell. Results:The expression of protein PPARγ existed in JEG-3 nutrient cell line. After the treatment of PPARγemotional ligand, the number of infiltrating membrance permeating cell decreased obviously. Compared with the contrast the difference was notable and existed dose-dependent relation. Conclusion: After being activated by its ligand 15-d-PGJ2, PPARγ inhibits the infiltrating ability of JEG-3 nutrient cells. This experiment provides theory and experimentation foundation for clinical application in the treatment of the PPARγ's ligand in abnormally infiltrating nutrient cell related pregnancy complication in the future.
