摘 要:目的建立不同展开体系中薄层色谱分析单糖或寡糖的有效方法。方法以淀粉寡糖、右旋糖酐寡糖以及古罗糖醛酸寡糖3个系列寡糖和半乳糖醛酸、葡萄糖醛酸、半乳糖、葡萄糖、阿拉伯糖、甘露糖、岩藻糖、木糖和鼠李糖9种单糖为研究对象,探讨其在不同展开体系下的薄层色谱行为。结果甲酸展开体系适合酸性寡糖和中性寡糖分离;氨水展开体系不适合于古罗糖醛酸寡糖分离,但有利于连接方式不同的中性寡糖的分离;三乙胺展开体系适合各种单糖的分离。除了2种糖醛酸外,其它7种单糖均显不同颜色。结论薄层色谱分析操作简便,适合于单糖或寡糖的快速分析。Purpose To establish an effective thin layer chromatography (TLC) method for the analysis of several oligosaccharides'and monosaccharides in different developing systems, Methods TLC with different developing systems was applied to the analysis of three series of oligosaccharides prepared from starch, dextran and guluronic acid, and also nine kinds of monosaccharides, that is, galacuronic acid, glucuronic acid, galactose, glucose, arabinose, mannose, fucose, xylose and rhamnose. Results Formic acid developing system is suitable for the analysis of acidic and neutral oligosaccharides. Ammonia developing system is not good for guluronic acid oligosaccharide analysis, but more suitable for the separation of neutral oligosaccharides with different linkage. Triethylamine developing system is suitable for different kinds of monosaccharides analysis. Except two kinds of uronic acid, other seven monosaccharides showed different color. Conclusion TLC is easy to operate and suitable for the rapid analysis of monosaccharides and oligosaccharides.
