摘  要:目的研究比较高乌甲素注射液和参附注射液对小鼠肝癌细胞株生长抑制及凋亡诱导作用。方法苔盼蓝计数,四甲基偶氮唑盐(MTT)法,流式细胞仪技术分析药物对细胞周期和凋亡指数的影响,琼脂糖电泳技术分析药物对DNA作用。结果氢溴酸高乌甲素注射液对H22作用、细胞数量与形态无明显影响;参附注射液可明显抑制小鼠肝癌H22的生长,细胞表现典型的凋亡形态,在浓度为50 mL/L时,出现明显的凋亡峰;琼脂糖凝胶电泳检测出现DNA ladder。结论参附注射液体外能抑制小鼠肝癌H22细胞株的增殖和诱导细胞凋亡,高乌甲素注射液体外对H22细胞的生长、凋亡没有影响。Purpose To investigate the effect of lappaconitine hydrobromide injection or Shenfu injection on murine hepatocarcinoma cell line H22 in vitro. Methods Trypan blue exclusion staining and MTF Method were used to detect cell proliferation. Cell cycle and apoptotic rate were detected by flow cytometry and agarose gel electrophoresis assay of DNA fragmentation. Results DNA ladder at the given concentration of lappaconitine hydrobromide injection showed no significant effect on the proliferation and morphology of H22, while Shenfu injection exhibited significant antiprolifcrative effect on H22. Typically apoptotic morphological changes and a typical hypodiploid peak were clearly observed in Shenfu injection-treated cells(50 mL/L group)and a ladder formation was also evident with an agarose gel electrophoresis of the extracted DNA. Conclusion Shenfu injection inhibits the proliferation of H22 and induces apoptosis in H22 while lappaconitine hydrobromide injection has insignificant effect on H22 in vitro.
