摘 要:在MS培养基中建立了连翘细胞悬浮培养,并研究了接种量、碳源、渗透胁迫及丙酮酸钠的影响.结果表明,接种量为90 g·FW·L^(-1)左右对连翘悬浮细胞生长和连翘甙合成有利,细胞的生长周期为18 d.碳源以蔗糖最合适,蔗糖浓度为30 g·L^(-1).渗透胁迫条件下以补加蔗糖和甘露醇最佳,细胞生长量和连翘甙的积累分别比对照增加了33.3%和89.9%.培养过程中添加20 mL 5 mmol·L^(-1)的丙酮酸钠对细胞生长和连翘甙的积累有促进作用.Cell suspension cultures were established from Forsythia suspensa (Thunb.) Vahl. callus cultures. The effect of several physiochemical factors on cell growth and phillyrin formation were studied. The results showed that the growth cycle of suspension cell were 18 d. Sucrose was better than glucose for the suspension cultures. The optimum concentration of sucrose for cell growth and flavonoid production was 30 g·L^-1. The medium with 5. 0 mmol/L sodium acetate promoted significantly the cell growth and phillyrin biosynthesis during the process of suspension cell cultures. Besides, it was a way of improving the phillyrin biosynthesis when the cells were treated with a manner of osmotic stress. Comparison with the control, the cell growth and phillyrin biosythesis have been increased by 33.3% and 89.9% resDectively.
