摘 要:目的观察分离提纯的牛α晶体蛋白(α-crystallin)对体外培养视网膜神经节细胞(retinal ganglion cells,RGCs)生长的作用。方法原代培养RGCs,Thy1.1免疫荧光法鉴定RGCs并进行纯度检测,不同浓度分离提纯的牛α晶体蛋白(10-7、10-6、10-5、10-4g/L)加入后,于3、12h、1、2d和3d在相差显微镜下观察RGCs形态学变化;计突起数目和轴突长度并与正常对照组、鼠α晶体蛋白组(10-4g/L)进行比较。结果分离提纯的牛α晶体蛋白组(10-4g/L和10-5g/L)RGCs突起数目及轴突长度显著大于正常对照组(P<0.05,P<0.01),牛α晶体蛋白10-4g/L与鼠α晶体蛋白组(10-4g/L)比较无明显区别(P>0.05)。结论分离提纯的牛α晶体蛋白(10-4g/L)能够促进RGCs生长,与鼠α晶体蛋白(10-4g/L)比较无明显区别。Objective To observe the effect of bovine α-crystallin on rat RGCs in vitro. Methods Rat RGCs were cultured and then identified by Thy1. 1 antibody. RGCs were treated with bovine α-crystallin at different concentration (10^-4, 10^-5, 10^-6, 10^-7 g/L) for 3 h, 12 h, 1 d, 2 d and 3 d, with the normal RGCs and RGCs treated with rat α-crystalline ( 10^-4 g/L) as controls. The changes of RGCs were observed, and the number and the length of outgrowing RGCs axons were calculated under microscope. Results The length and number of RGCs axons treated with 10^-4 g/L, 10^-5 g/L bovine α-crystallin, especially 10^-4 g/L, were more than those in normal control RGCs in the 1st, 2nd and 3rd day. The effect of 10^-4 g/L bovine α-crystallin had no difference from that of 10^-4g/L rat α-crystallin. Conclusion Bovine α-crystallin, especially of 10^-4 g/L, improves the survival of RGCs and the regeneration of axons, without difference with 10^-4 g/L rat α-crystallin on effect.
