摘  要:目的:观察骨髓间充质干细胞(MSCs)对博来霉素(BLM)引起的肺损伤大鼠肺组织胶原的影响,初步探讨其作用机制。方法:将SD大鼠MSCs异体注入受体大鼠体内,2周后观察肺组织病理学变化,同时检测肺组织羟脯氨酸含量及转化生长因子β1(TGFβ1)、血小板源性生长因子-A、B(PDGF-A,B)、胰岛素样生长因子-Ⅰ(IGF-Ⅰ)mRNA的变化。结果:MSCs的植入可使增生的大鼠肺泡间隔、基质胶原及Ⅰ、Ⅲ型胶原明显降低,使升高的肺组织羟脯氨酸及TGFβ1、PDGF-A,B、及IGF-ⅠmRNA的表达显著降低。结论:MSCs可减少肺损伤大鼠肺组织中的胶原含量,此作用与MSCs降低了多种促纤维化细胞因子的表达有关。Objective :To investigate the effect of bone marrow -derived mesenchymal stem cells (MSCs) on collagen of injured rat lung induced by bleomycin and its mechanism. Methods: MSCs, isolated from SD rats, were injected into recipient rats. Two weeks later, lung tissues were collected and lung structure, collagen Ⅰ ,Ⅲ and hydroxyproline were detected. The expression of TGFβ1, PDGF - A, PDGF - B and IGF - Ⅰ mRNA were also examined. Results: The thickness of alveolar wall, collagen Ⅰ ,Ⅲ and hydroxyproline in lung tissue in MSCs treatment group were reduced significantly than those in lung injury group. MSCs engraftment can also reduce the expression of TGFβ1, PDGF - A, PDGF - B and IGF - Ⅰ mRNA in injured lung. Conclusion : MSCs could reduce the collagen in injured lung and this effect may be associated with the decrease of TGF - β1, PDGF - A, PDGF - B and IGF- Ⅰ expressions induced by MSCs engraftment.
