摘 要:目的:探索形状(片层及条带)对构建工程化心肌组织(EHT)的影响.方法:原代心肌细胞与液态胶原,Matri-gel,2×DMEM混合,注入环形槽内形成心肌条带,注入方形槽内形成心肌片层,条带和片层培养1wk后行力学拉伸,继续培养1wk,行HE染色,鞣酸多彩(mason)染色,免疫组化染色以及激光共聚焦检测等对EHT进行评价.结果:拉伸前,心肌片层和心肌条带均可见点状自发性跳动,心肌片层拉伸后2d出现肉眼可见的一致性节律性跳动;心肌条带拉伸后4d形成肉眼可见的一致性跳动,14d后心肌条带仍然能维持长时间的跳动,形状和大小无明显改变,而心肌片层在去除力学拉伸后,发生明显的回缩,收缩力量减弱.HE染色,mason染色,免疫组化以及激光共聚焦检测结果显示,EHT拉伸后内部细胞均匀分布,沿拉伸方向伸展,细胞核完整,呈长梭形,含有丰富肌丝,与正常成年大鼠心肌组织类似.结论:片层和条带对构建EHT各有利弊.心肌片层易于构建,容易移植,但构建后生理性能不稳定,而心肌条带构建后生理性能较稳定,但构建成功率低.AIM : To sudy the effect of shape ( patch and ring) on the construction of engineered heart tissue (EHT). METHODS: Liquid collagen type I, Matrigel and 2 x DMEM were mixed with neonatal rat cardiac myocytes. The mixture was then infused into square molds to construct cardiac patches or infused into circular molds to construct cardiac rings. Seven days later, the EHTs were subjected to dynamic stretch for another 7 d. Light microscopy and transmission electron microscopy, routine HE staining, Mason's three color staining and immunohistochemical staining were used to analyze the EHTs. RESULTS: Spontaneous beating was seen in the EHTs before stretching. The engineered patches came to rhythmic synchronous beating at the 2nd day after stretching. The engineered cardiac rings finally came to synchronous beating at the 4th day after stretching. After 14 d, cardiac rings hadn't any changes, but cardiac patches reduced in size, and systolic force of cardiac patches dropped. HE staining, immunohistochemical staining and confocal laser scanning microscopy revealed that cardiac myocytes within EHT and construction of EHT resembled that of the native adult cardiac tissue in morphology and construction. CONCLUSION: Cardiac rings and cardiac patches have both advantages and disadvantages. Cardiac patche is easier to construct and transplant, but its physiological characteristics are unstable; cardiac ring's physiological characteristics are stable, but it's difficult to construct.
