摘 要:目的:探讨姜黄素对人肺腺癌A549/DDP细胞增殖活性的影响.方法:采用细胞计数法绘制亲代A549和A549/DDP细胞生长曲线,MTT法测定A549/DDP对顺铂的耐药指数以及姜黄素对A549/DDP细胞抑制率.将不同浓度姜黄素作用于A549/DDP细胞24h后,Hoechst33258染色荧光显微镜下观察细胞核形态,流式细胞仪检测细胞凋亡率.以20μmol/L的姜黄素作用细胞24h后,流式细胞仪分析细胞周期.结果:亲代A549细胞和A549/DDP细胞的生长曲线相似,倍增时间分别为(33.24±0.24)h和(32.88±0.11)h(P>0.05);顺铂处理A549和A549/DDP细胞72h后,半数抑制浓度(IC50)分别为13.57μmol/L和146.79μmol/L,A549/DDP细胞对顺铂的耐药指数为10.82.姜黄素对A549/DDP细胞增殖有抑制作用,并且呈时间浓度依赖性(P<0.05).姜黄素作用后,Hoechst染色显示A549/DDP细胞核缩小变形,致密浓染荧光成团块分布.流式细胞仪分析结果示细胞凋亡率呈明显剂量依赖关系(P<0.05).20μmol/L的姜黄素作用细胞24h,引发G2期阻滞.结论:姜黄素对人肺腺癌A549/DDP细胞增殖具有抑制作用,并呈时间、浓度依赖.其作用可能是通过阻滞细胞周期进程和诱导细胞凋亡来实现的.AIM: To explore the effect of curcumin on proliferation of human lung adenocarcinoma cell line A549/DDP. METHODS: Cell counting was used to plot the cell growth curves of A549 cells and A549/DDP cells. The resistance index of A549/DDP cells to cisplatin and inhibition rate of A549/DDP cells in different concentrations of curcumin were measured by MTT assay. After the cells were treated with curcumin, cellular morphology was observed under fluorescence microscope. Cell cycle analysis and apoptosis rate were detected by flow cytometry. RESULTS: The cell growth curve of A549 cells was similar to that of A549/DDP cells. The doubling generation time of A549 cells and A549/DDP cells were (33.24 ±0.24) h and (32.88 ± 0.11 ) h, respectively ( P 〉 0.05 ). After 72 h of treatment with cisplatine, ICso of cisplatine to A549 cells and A549/DDP cells were 13.57 μmol/L and 146.79 μmol/L, respectively, with the resistance index being 10.82. The inhibitory effect of curcumin on A549/DDP cells was shown in the dose-dependent and timedependent manners (P 〈 0.05 ). Under fluorescence microscope, cells treated with curcumin showed shrunk or distorted nucleus with fluorescence conglomerated. The apoptosis rate of A549/ DDP cells induced by curcumin was remarkably higher than that of the control cells, and cell cycle analysis showed G2 phase arrest. CONCLUSION: Curcumin exerts inhibitory effects on proliferation of A549/DDP cells in dose-dependent and timedependent manners possibly through inducing cell cycle arrest and cell apoptosis.
