摘  要:目的研究川芎嗪对癫痫的作用及其与白细胞介素-1受体(IL-1R)的关系。方法用腹腔注射戊四氮制作大鼠癫痫模型,致痫前后分组分别给予川芎嗪,观察大鼠出现癫痫发作的潜伏期及持续时间,并用脑电图仪和免疫细胞化学方法研究川芎嗪对癫痫大鼠脑电图和皮层及海马区IL-1R免疫反应阳性细胞的影响。结果对照组无癫痫发作。致痫前给予川芎嗪组大鼠较癫痫组大鼠癫痫发作潜伏期延长,持续时间缩短,差异有显著意义(P<0.01),脑电图高电位痫样波发放减少,大脑皮层和海马IL-1R免疫反应阳性细胞数减少,差异有显著意义(P<0.01)。致痫后给予川芎嗪组,上述指标与癫痫组相比,差异无显著意义(P>0.05)。结论川芎嗪能抑制癫痫发作,其作用机制可能与通过白细胞介素-1对免疫-神经的调节有关。Objective To study the effects of tetramethylpyrazine(TMP) in treating epilepsy and its relationship to interleukin - 1 receptor, Methods The epilepsy model of rats was induced by intraperitoneal pentylenetetrazol(PTZ) injection before and after tetramethylpyrazine (TMP) was administered respectively, The latency and continuous time of epilepsy rats were observed. The influence of TMP on eletroencephalogram(EEG) and the expression of interleukin-1 receptor β in cortex and hippocampus were also investigated by means of EEG and immunocytochemistry. Results TMP could decrease the activities of GPT and GOT in serum and the MDA , XOD , NOS and iNOS content of liver tissue , and increase the activities of GSH - PX content of liver tissue. In vitro, it indicated that TMP had a notable protective effects on the hepatocyte injured by CCl4 and could suppress the level of GPT. Conclusion The action of TMP to suppress epilepsy discharge may have relationship to its regulation on immune - nervous affect by the expression of IL - 1R.
