摘  要:目的探讨同种异体血管移植后肿瘤坏死因子(TNF)在移植的血管和心肌组织中的表达。方法用成年雄性新西兰大白兔做血管供者,日本大白兔做血管受者,进行颈动脉血管移植。通过新鲜植入、抗生素处理及液氮冷冻等方法对移植的血管进行处理,HE染色观察移植血管和心肌组织的形态学改变;免疫组织化学方法检测移植血管和受者心肌组织TNF表达的变化。结果TNF表达率在液氮冷冻组低于新鲜移植组和抗生素处理组,与自体原位移植组相似。新鲜移植组TNF表达率最高,各组间比较差异有显著性(P<0.05)。血管移植后,受者心肌组织的TNF的表达与移植血管的相似:新鲜移植组和抗生素处理组的TNF表达率较高,液氮冷冻组的TNF表达率较低(P<0.05)。结论同种异体血管移植能激发移植血管和心肌组织产生TNF,液氮冷冻处理可降低移植血管和受者心肌组织的TNF的产生。[Objective] To study the variance of tumor necrosis factor (TNF) in grafted blood vessels and myocardium. [Methods] The carotid vasotransplantation was performed with male mature Japanese albino rabbits as vascular recipients and New Zealand rabbits as vascular donors. Immunohistochemical methods were used to detect TNF expression in grafted blood vessels and myocardium. [Results] TNF positive expression was lower in the autoorthotopic transplantation group and the liquid nitrogen frozen group than that in the fresh transplantation group and the antibiotic-treated group. And as compared with the other groups, the positive expression of TNF was the highest in the fresh transplantation group, and there was statistic significance (P 〈0.05). After vasotransplantation, the findings of myocardium were allogenic as vasotransplantation. TNF expression was higher in fresh transplantation group than that in the other groups. Compared with auto-orthotopic transplantation group and liquid nitrogen frozen group, there was statistical significance (P 〈0.05). However, compared with antibiotic-treated group, there was no statistic significance. [ Conclusions] TNF expression of myocardium and transplanted vessel may be depressed by pre-treatment with liquid nitrogen frozen. Ischemia reperfusion injures and/or immunological rejection may stimulate the production of TNF in grafted blood vessels and many organs and induce the pathological lesions of grafted vasculature.
