摘  要:以禽网状内皮组织增生病病毒(reticuloendotheliosis virus,REV)的单克隆抗体11B154作一抗,建立了在冰冻切片中快速检测REV的免疫酶(IHC)和免疫荧光技术(Mc-IFA)。运用两种技术对人工接种REV的肉种鸡和疑似REV田间病例的肝脏、脾脏、腺胃、心脏、骨髓等器官的组织进行了检测,并同病毒分离方法进行了敏感性和特异性的比较。结果表明:在人工接种的肉种鸡中,采用IHC、Mc-IFA和病毒分离方法结果一致,均为阳性;在送检的疑似RE的24份病料中,用IHC检测出阳性率51.7%(13/24),Mc-IFA检测出阳性率62.5%(15/24),其中13份存在交叉;病毒分离的阳性率为37.5%(9/24),阳性数包括在13份IHC阳性病料中。由此表明冰冻切片免疫酶和免疫荧光技术都可用于临床快速检测,其中免疫荧光可用于早期的粗检,而免疫酶可用于细胞内抗原定位。An immunofluorescence and immunoenzyme assay mediated by monoclonal antibody 11B154 was developed to detect antigen of reticuloendotheliosis virus in froze histological section directly. Liver, spleen, proventriculus, heart and bone marrow were detected by the two methods and the sensibility and specificity were compared between Mc-IFA, IHC and virus isolation method on chicken, which were inoculated with REV strain or suspicious field samples. The results showed that the positive detection was observed by three methods when inoculated chickens were detected. Positive rate of field samples detected by Mc-IFA,IHC and viral isolation was 62.5 % (15/24) 51.6 % (13/24) and 37.5 % (9/24) ,respectively. 13 samples positive samples detected by Mc-IFA and IHC were same. The results indicated that both Mc- IFA and IHC could be used to detect antigen of REV quickly. Mc-IFA was higher than IHC in sensibility and lower than IHC in specificity while these two methods were higher than virus isolation ,in insensibility and specificity and could be performed to detect and identify.
