PCR detection of CAV and ALV in the embryonic chickens and 1-day old broiler chicks from AA broiler breeders

摘  要:应用PCR方法,对来源于山东省3个不同AA肉种鸡场的鸡胚、1日龄雏鸡中的CAV和ALV进行检测,从而了解CAV和ALV在此阶段对肉鸡的感染情况。试验直接采集样品的肝脏、胸腺、骨髓、脾脏、肾脏、法氏囊等组织提取DNA,进行PCR扩增及PCR产物的克隆和序列测定。结果显示,被检的3个肉种鸡场均有这两种病毒的阳性检出,其中CAV的阳性率是20.18%;ALV的阳性率13.49%,二者共感染的阳性率是6.24%。而且感染鸡各组织含量也有所差异,其中CAV以脾脏的最多,ALV以肾脏最多。同时对肝脏进行细菌分离培养时发现存在大肠杆菌、沙门氏菌和霉形体的混合感染。由此可见,在鸡胚和雏鸡体内存在CAV、ALV的感染以及与细菌性疾病的共感染。CAV( Chicken infectious anemia Virus) and ALV (Avian leukosis virus) were detected by PCR from embryonic chickens and one-day chickens that were collected from different 3 AA broiler breeder farms of Shandong province,we tried to see infections of CAV and ALV in this stage. DNA from liver, thymus, marrow, spleen, kidney, bursa fabricii were extracted and amplified by PCR, then evaluated by PCR. Cloned plasmid was sequenced. The results showed that CAV and ALV were proved to be positive in the three farms investigated, and the positive ratio of CAV and ALV were 20.18 % and 13.49 %. The co-infection ratio of ALV and CLV was 6.24 %. Positive ratio differed in different organs,and the highest positive ratio of CAV and ALV appeared in spleen,kidney,respectively. Meanwhile,we also found co-infection of Escherichla coli ,Salmonella and Mycoplasma in liver. The above results demonstrated that CAV and ALV as well as some bacteria co-infected in embryonic chickens and one-day chickens.
