摘  要:以丰花月季品种"红帽子"叶片为试材,研究了不同植物生长调节剂及暗培养时间对月季体胚诱导和植株再生的影响。结果表明,在5 mg/L 2,4-D的MS培养基上光下培养,20 d的愈伤组织诱导效果最好,诱导率达98%;在MS+1.2 mg/L TDZ+0.1 mg/L NAA+0.1 mg/L的培养基上暗培养8 d后转入光下培养,体胚诱导率为33.3%,植株再生率为16.7%,暗培养时间超过8 d,外植体易形成愈伤组织而不利于体胚的分化。Effects of combinations of plant growth regulators and induction period in the dark on somatic embryogenesis induction and plant regeneration of leaflet in Rosa chinensis rodhatte were studied. The results showed that after 20 days of induction on MS medium with 5 mg/L 2,4-D, the best callus was obtained, the induction frequency of callus was 99% ; after 8 days of induction on MS medium with 0.5 mg/L TDZ,0.01 mg/L NAA and 0. 1 mg/L GA3 in the dark transfering in the light, the highest plant regeneration was ob- tained. Induction of somatic embryogenesis and the regeneration rate were 33.3% and 16.7% , respectively. The time longer than 8 days of induction in the dark caused callus formation and the regeneration frequency was significantly reduced.
