摘 要:目的:考察高效毛细管电泳法(HPCE)在甲硝唑定量分析中的应用。方法:通过优化电泳条件,采用HPCE区带电泳法(CZE)对甲倍霜中甲硝唑进行含量测定。以40mmol/L Tris—H3PO4为缓冲液,电动进样10kV×20S,分离电压为+24kV,紫外300nm检测,分离柱温25℃。结果:以HPCE法测定甲硝唑对照品的保留时间为:8.17min;标准曲线为:Y=0.0394x+0.0078(r=0.9995);线形范围为:1.25~75.00μg/mL;批内RSD为:1.35%~2.01%,批间RSD为:3.11%~4.62%;甲硝唑平均含量为标示量的97.30%。结论:HPCE法取样量小,快速方便,结果可靠,可用于甲倍霜的含量测定。Objective To determine content of metronidazole (MTZ) in metronidazole beclometasone cream by high performance capillary electrophoresis (HPCE). Method Capillary zone electrophoresis (CZE) optimized previously was performed with electrokinetic injection time 20 seconds( 10 kV) at the separation voltage of 24 kV,detection wave length 300nm, and column temperature 25℃ with 40mmol/L Tris - H3PO4 solution as running buffer. Results The retention time of MTZ by HPCE was 8.17min. The calibration curve was y = 0.0394x + 0.0078 ( r = 0. 9995 ) with the content range of 1.25 - 75.00μg/mL. The internal day precision RSD was 1.35 -2.01% ( n =5) while the intraday precision RSD was 3.11-4.62% in =5). The mean content of MTZ in MBC by HPCE was 97.30%. Conclusion HPCE may be potential method in the concentration determination of metronidazole, because of its minute sampling, rapidness, convenience and accuracy.
