摘  要:目的观察胰岛素样生长因子-1(IGF-1)R抗体体内局部干预对动脉粥样硬化大鼠血管损伤后新生内膜增生的抑制作用。方法建立动脉粥样硬化大鼠颈动脉球囊损伤模型,随机分成三组。损伤术后,分别将缓冲液、对照抗体和IGF-1R抗体作用于大鼠损伤血管壁,观察三者对术后不同时期新生内膜增生、细胞增殖的影响,以及对血管平滑肌细胞功能状态的影响。结果经IGF-1R抗体干预后,与给予缓冲液和对照抗体干预相比:各时期内膜/中膜面积比显著降低(P<0.01);损伤后早期,VSMC增殖率明显下降(P<0.01);损伤后期,电镜下可见到较多的凋亡细胞。结论IGF-1R抗体可以抑制损伤后新生内膜增生,减少再狭窄发生率,并可抑制损伤后早期VSMC的增殖。Objective To observe the inhibition effect of IGF-1R monoclonal antibody on neointima hyperplasia after balloon injury in atherosclerosis rat. Methods 72 male wistar rats were used in this study. They were divided into three groups. Immediately after angioplasty, IGF-1R monoclonal antibody or control antibody or PBS was delivered into the injured segmet of the common carotid artery for local therapy. And then intimal-media ratio and ceil proliferation indices were measured at different time. Results Intimal-media ratio was signigicantly reduced(P 〈0.01) at each time in rats given IGF-1R antibody compared with those that received control antibody or PBS. At the beginning after injury, there was an obvious decrease in VSMC proliferation(P 〈0.01). We can also see a lot of apoptotic cells with electron microscope in 1 month to 3 months after injury in the rats given IGF-1R antibody. Conclusion IGF-1R monoclonal antibody can strongly inhibit neointima hyperplasia after balloon injury in atherosclerosis rats and it mainly inhibits VSMC proliferation.
