摘 要:根据已报道的Hoxc9基因序列保守区设计3对引物,利用PCR技术从安哥拉山羊血液基因组DNA中扩增Hoxc9基因序列。目的片段纯化后连接到PMD19-T载体上,经鉴定得到重组质粒。测序结果通过与Gen-Bank数据库中序列比对分析,确定该序列为Hoxc9基因,核苷酸序列长3224 bp,包括1个内含子和2个外显子,cDNA序列长783 bp,编码260个氨基酸。与哺乳动物氨基酸序列的同源性非常高,达到99.2%以上,同斑马鱼和日本鳉的同源性较低。Three pairs of primers were designed according to the conservative sequences of announced Hoxc9 gene. The Hoxc9 gene sequence of Angora goat was amplified by PCR. The PCR products were purified and then cloned into PMD19 -T vector. The recombinant plasmids were sequenced. By using the program of BLAST on database of GenBank , the sequence was identified as Hoxc9 gene. Its lengths of DNA and cDNA were 3 224 bp and 783 bp respectively, which encoded 260 amino acid. The amino acid sequence homology compared with that of other mammalians was more than 99.2% and was lower compared with that of other zebrafish and Japanese medaka.
