摘  要:目的:研究植酸对SGC-7901细胞的凋亡诱导作用及其机制。方法:采用MTT实验法观察植酸对SGC-7901细胞的增殖抑制作用;DNA琼脂糖凝胶电泳实验(DNA Ladder)观察植酸对SGC-7901细胞的凋亡诱导作用;Western blotting检测凋亡相关蛋白Fas、Bax及caspase-3的表达。结果:MTT实验结果显示,植酸对SGC-7901细胞具有生长抑制作用,并呈现剂量与时间的效应关系;经3.5mmol/L植酸处理72h的细胞出现了典型的DNA片断的梯形条带;植酸能够诱导caspase-3的活化,植酸处理组细胞中Fas、Bax蛋白比对照组表达增加,且呈现剂量-反应关系。结论:植酸对SGC-7901细胞具有诱导凋亡作用,Fas、Bax及caspase-3参与了植酸对人胃癌SGC-7901细胞的凋亡诱导作用机制。Objective: To explore the effect of phytic acid on apoptosis of SGC-7901 cells and its mechanisms. Method: The inhibiting action of phytic acid on SGC-7901 cells was examed by MTT assay, and the effect on apoptosis of SGC-7901 cells was examined by DNA gel electrophoresis, and the expressions of Fas, Bax and caspase-3 protein were detected by Western blot method. Results: From MTT assay, we observed that phytic acid inhibited the growth of SGC-7901 cells in dose and time dependent manner. Typical DNA ladder was found during gel eletrophoresis. Caspase-3 can be activated by phytic acid and the expressions of Bax and Fas protein in phytic acid group were higher compared with control group, and increased in a dOSekdependent manner. Conclusion: Phytic acid can induce apoptosis of SGC-7901 cells which may be correlated to apoptosis associated protein Fas, Bax and caspase-3.
