摘 要:目的观察土贝母皂甙对鸭乙型肝炎病毒的影响。方法用DHBV-DNA阳性雏鸭做动物模型,土贝母皂甙体内用量为1mg·kg-1·d-1、0.1mg·kg-1·d-1、0.01mg·kg-1·d-1,口服给药,每天1次,连续给药10天。采用鸭肝组织HE染色法和用药前后血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)检测药物对肝脏的毒性作用,用斑点杂交试验检测药物对雏鸭血清中DHBV-DNA的影响。同时以拉米夫定(2mg·kg-1·d-1)及生理盐水为阳性和病毒对照组。结果土贝母皂甙大剂量组(1mg·kg-1·d-1)ALT、AST在用药期间有所降低,说明土贝母皂甙在一定浓度下可能具有保肝作用。观察肝脏HE染色病理切片,各浓度组均未引起肝细胞明显的病理损害。斑点杂交试验发现土贝母皂甙大剂量组(1mg·kg-1·d-1)及拉米夫定组(2mg·kg-1·d-1)对血清中的DHBV-DNA都有明显的抑制作用,与本组用药前比较均有明显差异(P<0.01),但停药3d后各组DHBV-DNA未见转阴者,病毒复制均有反弹现象,说明两种药物在实验给药时间内不能彻底清除病毒血症;而土贝母皂甙中、小剂量在实验给药时间内对DHBV-DNA无抑制作用。结论土贝母皂甙可降低鸭血清中DHBV-DNA水平。Objective To observe the influences of tubeimoside on duck hepatitis B virus (DHBV). Methods DHBV- positive ducks were used as the hepatitis B animal models, and were treated with Tubeimoside orally at the dosages of 1mg·kg^-1·d^-1,0.1mg·kg^-1·d^-1.0.01 mg·kg^-1·d^-1 , once a day for 10 days. The toxicity of medicine in serum and liver of DHBV infected ducks were detected by alanine aminotransferase (ALT),aspartate aminotransferase (AST). Serum DHBV-DNA was detected by dot blot hybridization. At the same time, 3TC (2mg·kg^-1·d^-1) and the normal saline acted as the positive and negative control group respectively. Results The DHBV-positive ducks were respectively medicated with the three dose of Tubeimoside after 10 days, the results demonstrated that the dose of 1mg·kg^-1·d^-1 of Tubeimoside had possibly the hepatoprotective effect, its ALT and AST were obviously lower than that before treatment. In the experiment of dot blot hybridization, the group of Tubeimoside (1mg·kg^-1·d^-1)and 3TC (2mg·kg^-1·d^-1) could inhibit the replication of the serum DHBV DNA, it was obviously contrast compared with before treatment (P〈0,01). After cessation of the treatment for three days, DHBV-DNA replication in serum rebounded. It implied that the two kinds of medicines can not clean up the viremia thoroughly. In the dosages of 0.1mg·kg^-1·d^-1 and 0.01mg·kg^-1·d^-1 of Tubeimoside and the saline treated DHBV infected ducks, no changes of DHBV-DNA in serum were observed (P〉0.05). Conclusins In vivo, Tubeimoside can decrease serum DHBV-DNA level, but can not eliminate the viremia.
