摘 要:目的研究匹格列酮对正常人和甲状腺相关眼病(thyroid-associated ophthalmolpathy,TAO)患者眼眶前脂肪细胞增殖和分化的影响。方法培养正常人和TAO患者眼眶前脂肪细胞,在其增殖和分化的过程中加入匹格列酮。MTT法观察匹格列酮对前脂肪细胞增殖的影响,油红O染色定量的方法观察匹格列酮对前脂肪细胞分化的影响。结果浓度为1μmol.L-1的匹格列酮能够促进眼眶前脂肪细胞的增殖。在各个实验时间段,匹格列酮对眼眶前脂肪细胞有促分化作用,在48 h时作用最明显。与正常人比较,匹格列酮对TAO患者眼眶前脂肪细胞的促分化作用更明显。结论匹格列酮可能通过增加成熟脂肪细胞的数量改善胰岛素抵抗。对伴有糖尿病的TAO患者,匹格列酮治疗可能诱发眼部病变的加重。Objective To determine the effect of pioglitazone on the proliferation and differentiation of orbit preadipocytes in healthy adults and patients with thyroid-associated ophthalmopathy (TAO). Methods Pioglitazone was added to the culture media of orbit preadipocytes. The effects of pioglitazone on preadipocyte proliferation were examined by MTT. The effects on preadipocyte diferentiation were examined by oil red O staining. Results Pioglitazone of 1 μmol · L^-1 stimulated the proliferation of human orbit preadipocyte, and at each recorded time, human orbit preadipocyte differentiation was stimulated. The most obvious effect of stimulating differentiation was observed at 48. Pioglitazone could stimulate the orbit preadipocyte differentiation in patients with TAO more obviously than in healthy adults. Conclusion Pioglitazone may improve insulin resistance by increasing the number of mature adipocytes. It may aggravate eye disease in diabetic patients with TAO.
