摘 要:目的探讨干扰素-γ(IFN-γ)对正常人精子顶体酶(acrosin)活力的影响及其作用机制。方法采用BAEE/ADH联合法测定精子顶体酶活力,用高效液相色谱(HPLC)测定一氧化氮(NO),用生化法测定超氧化物歧化酶(SOD)和一氧化氮合酶(NOS)。用定磷法测定Na+-K+-ATPase和Ca2+-ATPase活性。结果IFN-γ可降低精子顶体酶活力(P<0.05),明显降低精子中Na+-K+-ATPase、Ca2+-ATPase和SOD活性(P<0.001);但显著提高精子NOS活性和NO浓度(P<0.001)。结论IFN-γ可降低精子顶体酶活力,其机制可能与精子中Na+-K+-ATPase、Ca2+-AT-Pase、SOD和NOS活性以及NO浓度等因素有一定关系。Objective To investigate the effect of interferon-gamma (IFN-γ) on the activity of acrosin in human sperm and its mechanism. Methods The acrosin activity was detected by BAEE/ADH method. The nitric oxide (NO) concentration was tested by HPLC method and the activity of superoxide dismutase (SOD), nitric oxide synthase (NOS), Na^+ -K^+ -ATPase, Ca^2+ -ATPase were tested by the biochemical technique. Results IFN-γ, decreased the activity of acrosin (P 〈 0.05) and the activities of SOD, Na^+ -K^+ -ATPase, and Ca^2+- ATPase (P 〈 0.001 ), but increased the NOS activity and the NO concentration in human sperm. Conclusion IFN-γ has some inhibitory effect on sperm acrosin activity, which could be attribute to their influence on activities of Na^+ -K^+ -ATPase, Ca^2+ -ATPase SOD, NOS, the NO concentration and so on.
