摘  要:目的:不同剂量链脲菌素(STZ)1次注射诱导小鼠糖尿病模型的建立,了解糖尿病发病机理,以及糖尿病小鼠的生殖能力。方法:ICR雌性小鼠随机分为正常对照组及200、150 mg.kg-1 STZ组。腹腔注射不同剂量STZ诱导小鼠糖尿病作为动物模型。葡萄糖测定试纸和尿液分析试纸条联合检测小鼠血糖和尿糖的变化,光镜观察胰岛的组织学改变情况,放射免疫方法检测小鼠血清中胰岛素的水平,并将制作的模型小鼠与正常雄鼠交配。结果:对照组血糖基本无变化,模型组血糖值随时间增加而增加,3周后稳定。高剂量组,血清葡萄糖浓度明显升高(P<0.001),血清胰岛素下降极显著(P<0.001);胰岛破坏严重。低剂量组分小鼠血清葡萄糖升高和未升高组,但血清胰岛素浓度均明显降低(P<0.01),胰岛内可见炎性细胞浸润。血糖升高小鼠交配能力下降,仅为46.9%,流产率73.9%,畸形率为9%,仔鼠出生死亡率20%。结论:不同剂量的STZ均能诱发糖尿病模型,且影响雌鼠的交配能力和子代的生长发育。[Objective] To establish type 1 diabetes mellitus (T1DM) induced by different doses of streptozotocin (STZ) and to observe their mating ability. [Methods] ICR mice were divided into control group and 200,150 mg·kg^-1 STZ treated groups. Animal models of diabetes mellitus were induced by different doses of STZ through intra peritoneal injection. Blood and urine glucose levels were detected by glucose and urine analyzing papers respectively. Pancreatic histological changes of islets were observed under light microscope. The levels of concentration of serum insulin were detected by radio-immunity kit And the diabetic mouse was mated to the normal male mouse. [Results] In the high STZ dose group, the concentration of serum glucose was significantly increased (P〈 0.001), the concentration of serum insulin was significantly decreased(P〈0.001). And pancreatic islet β cells were impaired. In the low STZ dose group, the concentration of serum insulin was decreased(P〈0.01). The proportions of mating, abortion,malformation and fetus death of treated group (Glucose〉18 mmol·L^-1) are 46.9%,73.9% ,9% and 20% respectively.[Conclusion] Different doses of STZ can induce diabetes mellitus, and can affect the female mouse mating ability and fetal development.
