摘  要:目的体外研究大鼠骨髓间充质干细胞(rMSCs)分化为血管内皮细胞的潜能。方法采用密度梯度离心法分离rMSCs,体外扩增并进行鉴定。在内皮细胞生长培养基(EGM-2)中以血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)定向诱导rMSCs向血管内皮细胞分化,观察细胞形态学变化。采用免疫细胞化学法和Dil荧光标记法,分别检测诱导后rMSCs的内皮细胞表型表达及对乙酰化低密度脂蛋白(Ac-LDL)的摄取能力。结果形态学观察显示,诱导后的rMSCs可见类似内皮细胞样改变,并表达血管内皮细胞特异的表面标志vWF和CD31,但仅少数细胞具有摄取Dil-Ac-LDL的能力。结论VEGF和bFGF在体外能够诱导rMSCs出现内皮细胞表型,但尚不完全具备成熟内皮细胞的功能特性。rMSCs具有向血管内皮细胞分化的潜能。Objective Study the differentiation of rat bone marrow-derived mesenchymal stem cells (rMSCs) into vascular endothelial cells in vitro. Methods The rMSCs were isolated by density gradient centrifugation and amplificated and identificated in vitro. Vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) within endothelial cell growth medium (EGM-2) were used to induce rMSCs differentiating into vascular endothelial cells. Morphological changes were observed. The induced rMSCs were detected by immunocytochemical and flow cytometry to find whether they had endothelial cell phenotypes, DiI-Ac-LDL uptake experiment was used to find whether the induced cells had mature endothelial cells function, Results In cell morphology, the induced rMSCs transformed into endothelial-like and expressed vascular endothelial cell-specific surface markers such as vWF and CD31. But few cells had ability in DiI-Ac-LDL uptake, Conclusion The rMSCs can obtain endothelial cell phenotypes after induced by YEGF and bFGF in vitro. However, the induced endothelial-like cells don't have completely mature endothelial cell functional properties. The rMSCs have potential to differentiate into vascular endothelial cells.
