摘  要:目的:以高效液相色谱法测定二仙汤中知母黄酮含量及以比色法测定知母皂苷含量。方法:HPLC法测定采用C18-ODS反相色谱柱,流动相:乙腈-25mmoL/LKH2PO(4pH=3.0)=1:9,检测波长:317nm,流速:0.8mL/min。比色法测定采用紫外可见分光光度计,检测波长:590nm。结果:知母新芒果苷、芒果苷分别在1.0μg/mL~100.0μg/mL及50.0μg/mL~1000.0μg/mL的浓度范围内呈良好的线性关系(r=0.9992,r=0.9997),回收率分别为97.656%、99.008%(RSD=1.54%,RSD=1.28%);知母总皂苷在8.0μg/mL~40.0μg/mL范围内呈良好的线性关系(r=0.9995),回收率为100.84%(RSD=1.29%)。结论:本方法简便、准确、灵敏度高、重现性好,可用于二仙汤的质量控制。Objective:To determine the content of Rhizoma Anemarrhenae flavones by HPLC and the content of Rhizoma Anemarrhenae saponins by colorimetric method in ER-XIAN-TANG.Methods:With HPLC method, the chromatobar was C18-ODS, the mobile phase was acetonitrile-25mmoL/L KH2PO4 (pH=3.0)=1:9, with the flow rate of 0.8mL/min and the detection wave at 317nm. UV-VIS spectrophotometer was used in colorimetric method and the detection wave was at 590nm.Results:A good linear relationship was obtained between the peak areas and the concentrations of neo-mangiferin and mangiferin respectively in the range from 1.0μg/ mL- 100.0μg/mL ( r=0.9992 ) and 50.0μg/mL- 1000.0μg/mL ( r=0.9997 ), the average recoveries were 97.656% ( RSD = 1.54% ) and 99.008%(RSD=1.28% ), respectively.As well the good linear relationship of Rhizoma Anemarrhenae saponins in the range from 8.0μg/mL-40.0μg/mL(r=0.9995 ) was obtained and the average recoveries were 100.84%(RSD=1.29% ).Condusion:This method is convenient, rapid, and brings about good recovery, it can be used for quality control of ER-XIAN-TANG.
