摘  要:紫草素及其衍生物是中药紫草的有效化学成份,文章对紫草素抗肿瘤作用及其机制的研究作一综述。紫草素能抑制多种肿瘤细胞生长增殖,致肿瘤细胞凋亡,其作用机制包括:调节MAPK信号通路、引起Bcl-2蛋白家族表达变化从而促进细胞色素C的释放、激活Caspase蛋白酶家族启动凋亡、致肿瘤细胞周期阻滞、影响死亡受体家族及其相关蛋白活性等方面。动物实验和临床观察表明:紫草素具有较强的抑癌作用,对正常细胞毒副作用小。Shikonin, an ingredient of Lithospermum erythronrhizon is a traditional Chinese medicine. In this review, the recent researches in antitumor activities and mechanisms of Shikonin and it's derivatives will be summarized. Shikonin can inhibit tumor cell growth, cell proliferation and induce apoptosis in a variety of cancer cell lines in vitro .The mechanisms include that shikonin adjusts the MAPK singnal pathway, regulates the expression of the Bcl-2 family proteins to promote the release of cytochrome c, induces the cell apoptosis by activating the Caspase family proteins, leads to the cell cycle arresting and adjusts the activity of the death receptor family and it's related proteins. The researches by animal test and clinical observation indicate that shikonin has strong antitumor activities with less side-effect to normal cell.
