摘　　要：目的：用表达hCGβ的重组乳酸杆菌活疫苗经口接种不同品系小鼠产生抗hCGβ免疫应答。方法：用10^8、10^9、10^10个重组乳酸杆菌Lb．hCGβ灌服6～8周龄雌性BALB／c和C57BL／6小鼠。3周后用相同剂量加强免疫1次。间接ELISA检测初次免疫后2～8周血清和阴道灌洗液抗hCGβIsG和IgA抗体滴度。流式细胞仪分析B和T细胞的增殖情况，ELISpot计数抗hCGβIsG和IsA抗体分泌细胞克隆数。结果：不同品系小鼠经口腔接种10^9、10^10Lb．hCGβ可诱导相近的免疫应答；而10。诱导的抗体滴度较低。在加强免疫后，10^9和10^10Lb．hCGβ诱导的抗血清结合hCGβ抗原的能力〉100ng／n11。Lb．hCGβ口腔接种后，BALB／c小鼠子宫和阴道的B细胞增殖；C57BL／6小鼠子宫及阴道的抗体分泌细胞克隆数较脾脏明显增多（P〈0．05）。结论：重组乳酸杆菌活疫苗Lb．hCGβ可经口接种诱导有效的抗hCGβ免疫应答，产生的应答强度与免疫剂量呈正相关。Objective：To induce an anti-hCGβ antibody response in reproductive tract and the peripheral by oral inoculation with a recombinant alive Lactobacilli expressing hCGβ in different strains of mouse. Methods ：The 10^8 , 10^9, 10^10 Lb. hCGβ, a recombinant Lactobacillus expressing hCGβ, were inoculated orally into 6-8-week-old female BALB/c and C57BL/6 mice. The immunized mice were all boosted with the same dose at interval of 3 weeks. An indirect ELISA was used to determine anti-hCGβ IgG and IgA antibodies in vaginal lavage and serum that were got at the end of 2-8 weeks after the primary immunization. The B and T lymphocytes of spleen, uterus and vagina were prepared respectively, and their proliferation was analyzed by flow cytometry. The numbers of the cells secreting anti-hCGβ IgG or IgA antibody were detected by ELISPOt. Results：Inoculation of 10^9 or 10^10 alive Lb. hCGβ could induce similar response while 10^8 induce a lower attitude of response. The antiserum of either 10^9 or 10^10 Lb. hCGβ immunization after booster could neutralized more than 100 ng/ml of hCG antigen both in BALB/c and C57BL/6 mice. The B cell proliferation of the reproductive mucosa was more significantly higher than that of spleen in BALB/c mice （ P 〈 0. 05 ）. The numbers of IgG or IgA antibody-secreting cells in uterus and vagina were higher significantly than in spleen of C57BL/6 mice（P 〈0. 05）. Conclusion：The Lb. hCGβ as a llve vaccine can induce an effective humoral responses in reproductive tract and the peripheral through oral inoculation in a pattern of dosedependent.
