摘  要:目的通过体外观察非洲臀果木提取物对由碱性成纤维细胞生长因子(bFGF)所致NIH3T3细胞增生的作用来了解其治疗良性前列腺增生的机理。方法在体外培养NIH3T3细胞后,分别在其中加入bFGF和非洲臀果木提取物。继续培养48h,然后每个样本取细胞悬浮液在光学显微镜下进行细胞观察,并用溴化甲基噻唑基四唑(MTT)法测定细胞OD值。最后比较各组细胞生长情况,并进行统计学分析。结果在NIH3T3细胞中加入bFGF后,细胞记数明显增多,再加入非洲臀果木提取物后,细胞记数明显减少。用MTT法测定细胞OD值,加入bFGF后,OD值明显增加,再加入非洲臀果木提取物后,吸光度(OD值)明显减少。以上结果有统计学意义。结论bFGF对NIH3T3细胞有明显刺激生长的作用。非洲臀果木提取物不论有无bFGF存在对NIH3T3细胞的生长均有抑制作用。Objective To observe the inhibitory effects of Pygeum africanum extract on proliferation of cultured 3T3 fibroblasts stimulated by basic fibroblast growth factor (BFGF) in order to gain an insight into its mechanism of action in the treatment of benign prostatic hyperplasia (BPH). Methods BFGF and Pygeum africanum extract were added to NH3T3 cells separately, and cultured for 48 h. The cell suspension was observed under an optic microscope, and the value of optic density (OD) was detected by MTT method. Results Addition of BFGF in NIH3T3 cells increased the number of cells and the OD value, and addition of Pygeum africanum decreased the number of cells and the OD value. The difference was statistically signifieant. Conclusions BFGF stimulated the growth of NIH3T3 cells, while Pygeum africanum extract inhibited the growth of NIH3T3 cells either in the presence or in the absence of BFGF.
