摘 要:目的:探讨湖南药用植物菝葜种质资源的亲缘关系。方法:以湖南金刚藤主产区的15个菝葜材料为研究对象,用RAPD技术对其进行亲缘关系分析。在建立适合菝葜植物的PCR-RAPD反应体系的基础上,从45个随机引物(10 m er)中筛选出14条,对所有供试材料进行正式扩增,共获得14张DNA指纹图谱,144条DNA谱带,其中有135条为多态带。建立了基于RAPD的菝葜植物亲缘关系树形图。结果:成功地将湖南金刚藤主产区的菝葜植物划分为明山类、会同类、靖州类、雪峰山类和鸡公界类的菝葜归为会雪类、靖鸡类和明山类。会雪类与靖鸡类与明山菝葜相比较,会雪类与靖鸡类的亲缘关系又较近。结论:本研究揭示了湖南省不同菝葜类型之间亲缘关系,这个研究结果是国内外首次对湖南不同菝葜类型之间亲缘关系的报道。Objective: To investigate the genetic relationship for medicinal plant germplasm resource of smilax in Hanan province. Methods: Fifteen Smilax materials in Hanan province were made as study objects, their genetic relationship were analyzed by random amplified polymorphic DNA markers(RAPD) technology. 14 primers were screened from 45 arbitrary 10 -mer primers, and total of 144 DNA bands were amplified, 135 of which (95.4%) were polymorphism. According to the data, a tree diagram of genetic relationship was constructed. Results : Smilax plants, which had been classified 5 groups, Ming mountain group, Huitong group, Jingzhou group, Xuefeng mountain group and Jigong mountain group, were divided into Huixue group, Jingji group and Ming mountain group. And Huixue group and Jingfi group took on intimate relationship among three groups. Conclusion: The studies indicate the genetic relationship in different patterns of smilax in Hunan province, that's first domestic report on this study.
