摘 要:目的探讨妊娠期慢性使用可卡因对新生兔肝脏、肾脏的影响。方法将24只怀孕的日本长耳白兔随机分为实验组和对照组,每组12只。两组均于怀孕15 d至分娩(30~31 d),分别从耳缘静脉注射可卡因(剂量为5 mg·kg^-1·d^-1)或0.9%氯化钠溶液(剂量为1 mL·kg^-1·d^-1)。检测血清中天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总蛋白(TP)、总胆红素(T-BiL)、直接胆红素(D-BiL)、清蛋白(ALB)、碱性磷酸酶(ALP)、总胆汁酸(TBA)、谷氨酰转肽酶(γ-GT)、尿素氮(BUN)、尿酸(UA)、血肌酐(Cr)、血糖(Glu)。肝细胞中细胞色素P450含量。结果实验组血清AST、ALT、γ-GT、TP、ALP、BUN、Glu均高于对照组(P〈0.01),血清ALB、D-BiL、TBA、Cr、UA均低于对照组(P〈0.01)。实验组P450含量高于对照组(P〈0.01)。结论妊娠期使用可卡因对新生兔肝脏、肾脏造成严重损害造成新生兔高胆红素血症。Objective To study the effects of chronic prenatal exposure to cocaine on liver and kidney in the newborn rabbits at birth. Methods The pregnant Japan long-eared white rabbits were divided randomly into experimental group (n = 12)and normal control group (n = 12). Pregnant rabbits were treated with cocaine hydrochloride 5 mg · kg^-1 or normal saline 1 mL · kg^-1 intravenously ,once a day from days 15 of pregnancy until days 30 or 31. Serum aspartate transaminase(AST) ,alanine transaminase (ALT) , gamma glutamyl-transferase (γ-GT ) , total protein ( TP ) , total bilirubin ( T-BiL ) , alkaline phosphatase (ALP) ,total bile acid(TBA) , direct bilirubin (D-BiL) , blood urea nitrogen(BUN) , creatinine(Cr) , uric acid (UA) , glucose (Glu) and cytochrome P450 were examined . Results The mean levels of serum AST, ALT, γ-GT, TP, ALP, BUN, T-BiL Glu were significandy higher in the experimental group than those in the control group(P 〈0.01 ). The mean levels of serum ALB,D-BiL, TBA, Cr, UA were significantly lower in the experimental group than those in the control group (P 〈 0.01 ). Cytochrome P450 contents were higher in experimental group than those in control group ( P 〈 0.01 ). Conclusion Cocaine application in pregnancy period causes severe injury on liver and kidney with hyperbilirubinemia in newborn rabbits.
