摘 要:利用原子力显微镜(AFM)对透射电镜生物制样的超薄切片进行扫描成像,研究细胞内的超微结构。对小鼠肝脏组织进行常规的透射电镜生物制样处理,Epon 812包埋聚合,钻石刀切片70nm厚超薄切片,移到新鲜解离的云母上。所得图像能够清晰分辨细胞和亚细胞结构,并获得细胞内亚细胞器的粘弹性等物理性质,为进一步研究细胞的结构和功能提供了新的技术方法。To observe the ultrastructure of mice hepatocyte by the techniques of uhrathin sections with Atomic force microscopy (AFM). The mice liver tissue was fixed with the 3% glutaraldehyde for 2h, and embedded in Epon 812. Then it was cut into 70nm thick uhrathin sections by diamond knife, which were then put onto the top of new cleaved sheets of fresh mica. The images of cellular and subcellular structures were clearly observed. Otherwise, we obtained the viscoelasticity of subcellular structures and other physical properties. AFM provides an effective method to study the structures and functions of ultrathin sections of liver tissue.
