摘　　要：目的:建立柱前衍生化 HPLC 法测定 O/W 型青蒿素脂肪乳剂中青蒿素含量的方法。方法:采用青蒿素的柱前碱性衍生反应,Agilent HC—C_(18)(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇:0.05 moL·L^(-1)醋酸钠-醋酸缓冲液(pH5.8)=66:34;流速:0.5 mL·min^(-1);柱温:25℃;检测波长:259.8 nm。结果:青蒿素的柱前衍生化反应彻底可靠,该方法在检测波长259.8 nm,试样浓度0.3～10μg·mL^(-1)范围内,与检测峰峰面积呈良好的线性关系(r=0.999),平均回收率为96.1%,RSD=1.1%。结论:本方法简便、精密,回收率与精密度高,重现性好,可用于青蒿素脂肪乳剂中青蒿素含量的测定。Objectives ：To establish a method to determine the content of artemisinin in oil/water fat emulsion by precolumn derivation HPLC. Methods：The column employed was Agilent HC -C18 （250 mm ×4. 6 mm,5 μm） ,the mobile phase was methanol：0. 01 mol . L^-1 NaAC -HAC buffer （pH 5.8） （66： 34） ,the flow rate was set at 0. 5 mL . min^ - 1, the column temperature was 25 ℃ and the wavelength was 259. 8 nm. Results ： The density of artemisinin had good linear correlation with absorbance in the range of 0. 3 -10 μg . mL^-1 （r = 0. 999） ,the average recovery was 96. 1% （ RSD = 1.1 ）. Conclusions： The present method is simple, precise and replicable resulting in high recovery and accuracy. It can be used to determine the content of total artemisinin in oil/water emulsion of artemisinin.
