摘  要:分别用0.9%NaCl水溶液、热水、0.5mol/LNaOH水溶液和88%甲酸从新鲜茯苓菌核体中提取出5种多糖:PC1、PC2、PC2-A、PC3和PC4.用IR、HPLC、GC、1HNMR和13CNMR等方法分析了它们的组成和结构.结果表明,PC1、PC2和PC2-A由D-葡萄糖、D-木糖、D-甘露糖和D-半乳糖组成,PC1和PC2-A含葡萄糖醛酸,其葡萄糖含量随提取过程的进行逐渐增加.PC3主要由线型β-(1→3)-D-葡聚糖组成,它占茯苓多糖的绝大部分.PC4由β-(1→3)-D-葡聚糖组成,含少量支链和葡萄糖醛酸.Five polysaccharides (PC1, PC2, PC2-A, PC3 and PC4) were isolated from a fresh sclerotium of Poria cocos by using 0. 9% NaCl aqueous solution, hot water, 0. 5 mol/L NaOH aqueous solution and 88% formic acid, respectively. The components and structures of the polysaccharides were analyzed by IR, GC, HPLC, 13C NMR and 1H NMR. The polysaccharides of PC1, PC2 and PC2-A consist of D-glucose, D-xylose, D-mannose and D-galactose, and the D-glucose content increases with the progress of isolation process. The PC1 and PC2-A contain glucuronic acid. The polysaccharide PC3 contains mainly linearβ-(1-3)-D-glucan, and is the major fraction of Poria cocos polysaccharides. The PC4 is com-posed of β-(1-3)-D-glucan with a few of braching and glucuronic acid.
