摘 要:从发病信鸽的体内分离到1株新城疫病毒(Newcastle disease virus,NDV)PB9601,经病毒蚀斑纯化后动物回归试验可产生典型的NDV病变,其鸡胚平均死亡时间(MDT)、一日龄雏鸡脑内接种的致病指数(ICPI)、六周龄鸡静脉接种的致病指数(IVPI)分别为50.5h、1.65和2.36,表明NDV为强毒。对其融合蛋白(F)和血凝素神经氨酸酶(HN)基因分别进行克隆测序,发现F蛋白多肽裂解位点的氨基酸基序为112 KRQKRF 117,符合鸽源NDV强毒序列。F基因分型及同源性比较显示:PB9601与YN-03、TW-98等鸽源毒株属于Ⅵ型,其F基因与疫苗株基因Ⅱ型LaSota的氨基酸同源性为88.6%,与疫苗株基因Ⅰ型V4的氨基酸同源性为91.0%,与传统强毒基因Ⅸ型F48E9的氨基酸同源性为90.6%;与国内鸽分离株GX-05、YN-03和TW-98的氨基酸同源性分别为92.1%、95.5%和98.6%;与国外鸽分离株Capital3-97、Tigre6-99、1166-00、IT-227-82和2736-00的同源性分别为93.3%、91.9%、94.6%、95.1%和93.6%。HN基因氨基酸同源比较显示:PB9601与LaSota、F48E9和V4的氨基酸同源性分别为87.2%、89.2%和87.6%;与国外鸽分离株1166-00、IT-227-82和2736-00的同源性为93.7%、95.5%和92.3%。PB9601与国内外鸽分离株、LaSota、F48E9和V4相比,具有明显的地域性和种属性。One Newcastle disease virus (NDV) isolate named PB9601 and collected from homing pigeon was characterized molecularly and biologically. And the isolate was determined as a virulent strain by the mean death time (MDT) of 50.5 h, the intmcerebral pathogenicity index (ICPI) of 1.65, and the intravenous pathogenicity index (IVPI) of 2.36. The fusion (F) and hemagglutinin-neuraminidase (HN) genes of the isolate PB9601 were cloned and sequenced. The deduced amino acid sequence of F gene showed that the virus shared a velogenic motif 112 KRQKRF 117 at the cleavage site. Phylogenetic analysis showed that the viruses derived from pigeon such as PB9601, YN-03 and TW-98 belonged to genotype. For the Fgene of the isolate PB9601, the homologies of the deduced amino acid sequences were 88.6%, 91.0% and 90.6%, respectively, compared with strains LaSota, V4 and F48E9; 92.1%, 95.5% and 98.6% individually for isolates GX-05, YN-03 and TW-98 from host pigeon in China, and 93.3%, 91.9%, 94.6%, 95.1% and 93.6% respectively for pigeon-origin isolates Capital3-97, Tigre6-99, 1166-00, IT-227-82 and 2736-00 overseas. By comparison of the deduced amino acid sequences of HN gene, the isolate PB9601 with the exotic pigeon-origin strains 1166-00, IT-227-82 and 2736-00 shared higher homologies of 93.7%, 95.5% and 92.3%, and lower homologies of 87.2%, 89.2% and 87.6% with LaSota, F48E9 and V4. The results indicated that PB9601 had some obviously regional and genus traits among NDV strains.
