电针刺足三里穴对内囊出血大鼠胃黏膜损伤的影响

摘　　要：目的探讨电针刺足三里穴对大鼠内囊出血致胃黏膜损伤的内在保护作用机制。方法雄性SD大鼠40只,随机分为空白对照(A)组、内囊出血(B)组、假手术(C)组、内囊出血+电针(D)组、假手术+电针(E)组,每组8只。将大鼠尾部血缓慢注射入左侧内囊后肢内建立大鼠内囊出血模型,利用G6805-A电针仪对大鼠进行针刺干预。采用放免分析法测定各组大鼠血浆ET的含量,用生化法测定NO和MDA的含量,并按照Guth标准计算各组溃疡指数(uI)。结果B组与A、C和E组比较,UI明显上升(P<0.01);而血浆NO水平下降(P<0.01),ET、MDA明显上升(P<0.01)。D组与B组比较,UI有所下降(P<0.05);同时血浆NO水平上升(P<0.05),ET、MDA水平下降(P<0.05)。A、C和E组各组之间比较,各指标无统计学意义。结论内囊出血可引起大鼠胃黏膜出现应激性溃疡。电针刺足三里穴对内囊出血致大鼠胃黏膜损伤具有保护作用,其机制可能与调节机体胃黏膜血流量、胃肠激素及氧自由基代谢的有关。Objective To investigate the protective effects of electroacupuncture at zusanli point on gastric mucosa after internal capsule hemorrhage in rats. Methods 40 male SD rats were randomly divided into 5 groups ： control group （ A ） , internal capsule hemorrhage （ ICH） group （ B ） , sham - operation （ SO） group （ C） , ICH ＋ electroacupuncture group （ D） and SO ＋ electroacupuncture group （ E）. Each group has 8 rats. After arterial blood from the rat tail artery was slowly injected into hindlimb of the left internal capsule, the capsular hemorrhage model was established in rats. The electric stimulation was from a medical EA apparatus （ Model G6805 - A ）. Radioactive - immunon - assay methods were used to measure the plasma levels of endothelin（ ET） , the level of nitric oxide（ NO） and malondialdehyde （ MDA） were measured by biochemical methods. The extent of gastric mucosal damage was evaluated by ulcer index（ UI） , according to the Guth exponential method. Results In the B group, the mean value of UI increased greatly（ P 〈 0.01 ） compared with A, C and E groups, at the same time, the plasm level of NO decreased（ P 〈0.01 ） , but the level of ET and MDA significantly increased（ P 〈0.01 ）. In the D group with administration of 3 days electroacupuncture, the UI decreased （ P 〈 0.05 ） , the level of NO increased （ P 〈 0. 05） ,but the level of ET and MDA decreased（ P 〈0.05）. No significant changes of UI and the plasm level of substance were found between the A, C and E groups. Conclusion The cerebra internal capsule hemorrhage administration can induced stress ulcer in rats. Electroacupuncture at zusanli point may play some important role on protcet gastric mucosa in internal capsule hemorrhage model rats. This function may be connected with its regulation with the changes of gastric mucosal blood flow, plasma levels of gut hormone and oxygen free radicals.
