摘 要:目的:观察金匮肾气丸不同极性部位及不同极性部位组合对氢化可的松致肾阳虚大鼠睾酮和氧化应激的影响,寻找金匮肾气丸处方中对肾阳虚大鼠有治疗作用的药效部位。方法:雄性SD大鼠ig氢化可的松50mg.kg-1,qd,给药10 d,造成肾阳虚模型。成模后次日,大鼠给予金匮肾气丸全方及其石油醚、氯仿、乙酸乙酯、正丁醇和水提部位的混悬液,药液浓度以生药量计均为0.5 g.mL-1,每日ig体积为10 mL.kg-1,qd,给药20 d;于末次给药后取血,测定血清睾酮、丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性。结果:与正常组比较,肾阳虚大鼠体重下降(P<0.01),睾酮含量和SOD活性降低(P<0.01),MDA水平升高(P<0.01)。金匮肾气丸及其水提部位能不同程度地改善肾阳虚大鼠的上述变化(P<0.01,P<0.05),金匮肾气丸的水提部位+正丁醇部位改善肾阳虚大鼠上述变化的作用更显著(P<0.01)。结论:金匮肾气丸水提部位能改善可的松致肾阳虚大鼠睾酮和氧化应激作用,正丁醇部位可增强药效,水提+正丁醇部位为金匮肾气丸治疗大鼠肾阳虚的主要药效部位。Objective: To determine the active fractions of Jinkuishenqiwa, we observed me effects of different polar fractions from the prescription of Jinkuishenqiwan and their combinations on testosterone and oxidative stress in rats with kidney-yang deficiency induced by hydrocortisone. Methods: Male SD rats were treated by ig hydrocortisone (50 mg·kg^-1·d^-1) for 10 days to induce kidney-yang deficiency. The fractions were extracted from Jinkuishenqiwan by ligarine, chloroform, ehtyl acetate, n-bu- tanol and water. From the 2rid day after hydrocortisone treatment, rats were treated with ig suspensions of Jinkuishenqiwan or its extractions ( each at mL·kg^-1 of solutions containing 0.5 g crude drug in 1 mL) once daily for 20 days. After the last administration, blood samples were collected, and the serum levels of testosterone, malonic dialdehyde (MDA) and superoxide dismutase (SOD) were determined, respectively. Results: The body weights, testosterone content and SOD activity of rats with kidney-yang deficiency were significantly decreased (P 〈 0.01 ) , whereas the level of MDA was significantly increased (P 〈 0. 01 ). Jinkuishenqiwan and its aqueous extracts remarkably improved the changes in rats with kid- ney-yang deficiency (P 〈 0.01 - 0.05) . Combination of water extracts with n-butanol extracts showed a more potent effect (P 〈0.01 ). Conclusions: Aqueous extracts of Jinkuishenqiwan improve the changes in testosterone level and oxidative stress of rats with kidney-yang deficiency induced by hydrocortisone; this effect can be enhanced by n-butanol extracts. These findings suggest that aqueous extracts and n-bu- tanol extracts may be the major active components of Jinkuishenqiwan.
