摘 要:目的:探讨腹部火器伤肠管穿透后NF-κB在肝细胞凋亡信号转导中的作用,了解腹部火器伤肠管穿透后继发性肝损伤机制。方法:健康长白仔猪42头随机等分为对照组和伤后1、2、4、8、12和24h组,建立腹部火器伤肠管穿透模型,用免疫组化图像分析法测定各组肝内NF-κB活性,观察肝细胞凋亡和肝功能变化情况。结果:伤后各组肝内NF-κB活性明显高于对照组,并于伤后1h和8h出现2个高峰(P<0.05)。肝细胞凋亡指数和血清ALT水平于伤后显著增高(P<0.05),并与NF-κB活性变化基本一致。结论:腹部火器伤肠管穿透后肝内NF-κB活性增强,肝细胞凋亡增多,导致肝损伤。Objective: To investigate the role of NF-κB in signal transduction of hepatocyte apoptosis in liver injury. Methodes A total of 42 Chang-Bai piglets were divided into 7 groups: control group, 1, 2, 4, 8, 12, and 24 hours wound group. The model of intestinal perforations due to abdominal firearm wound was established in wound groups. Hepatic NF-κB activity was measured with immunohistochemical staining and image analysis in all groups. Hepatocyte apoptosis indexes and serum ALT levels were also determined. Results: Levels of hepatic NF-κB activity in wounded groups were significantly elevated compared with control group, and there were two peaks (1 and 8 hours group P〈0.05). Hepatocyte apoptosis indexes and serum ALT levels were markedly in creased compared with control group(P〈0.05). All these changes were positive correlation with the changes of hepatic NF-κB activity. Conclusion: After intestinal perforations due to abdominal firearm wound, hepatic NF-κB activity increased and it might induce hepatocyte apoptosis and liver injury.
