摘  要:通过检测大鼠腹腔巨噬细胞(M)胞浆游离钙离子浓度([Ca2+]i)和45Ca内流与外排研究了不同浓度白芍总甙(TGP)对MCa2+跨膜转运的影响.结果表明,胞外有Ca2+时,TGP(2.5-20mgL-1)对亚适浓度卡西霉素(0.1μmolL-1),脂多糖(LPS,5mgL-1)和甲酰三肽(fMLP,1μmolL-1)升高M的[Ca2+]i具有明显的增强作用;而胞外无Ca2+时,TGP对LPS或fMLP刺激胞内钙池释放Ca2+引起[Ca2+]i升高的作用无明显影响;TGP可使卡西霉素刺激M的45Ca内流明显增加,同时较高浓度(10-40mgL-1)亦浓度依赖性促进45Ca外排.这些结果提示,TGP有促进胞外钙内流作用,较高浓度TGP可促进胞内钙外排,从而降低[Ca2+]i.Effect of different concentrations of total glucosides of paeony (TGP) on calcium ion across membrane of rat peritoneal macrophages (PM ) were studied by measuring cytoplasmic free Ca 2+ concentration ([Ca 2+ ] i) and 45 Ca fluxes in PM  from rats. The results showed that TGP (2.5-20 mg·L 1 ) had a marked enhancement on elevated [Ca 2+ ] i in PM  stimulated by the suboptimal concentrations of calcimycin(0.1 μmol·L 1 ), lipopolysaccharide (LPS, 5 mg·L 1 ) or formyl methionine leucine phenylalanine (fMLP, 1 μmol·L 1 ), in the presence of extracellular Ca 2+ (1 mmol·L 1 ), but TGP did not have any effect on increased [Ca 2+ ] i through releasing Ca 2+ in its internal reservoir of PM  induced by LPS or fMLP. TGP could increase 45 Ca influx, but its higher concentrations (10-40 mg·L 1 ) could promote 45 Ca efflux. These results suggest that TGP can increase the influx of extracellular calcium, and that the higher concentration of TGP can promote efflux of intracellular calcium and then down reglate the intracellular calcium concentration.
