胶原诱导关节炎大鼠滑膜中血管内皮生长因子和内皮抑素的表达及意义

摘  要:目的观察血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、内皮抑素(endostatin)在胶原诱导关节炎(CIA)大鼠不同时期滑膜中的蛋白表达,通过CD34给血管密度计数.明确生长因子在血管翳形成中的作用机制。方法建立CIA大鼠模型,测量关节体积,计算关节病理积分,并用免疫组织化学染色检测VEGF、bFGF、endostatin和CD34的蛋白表达。结果模型组大鼠滑膜VEGF和bFGF表达明显高于正常组大鼠(P〈0.01),并且二者表达存在相关性(P〈0.01),内皮抑素表达模型组与正常组比较差异无统计学意义(P〉0.05)。结论滑膜中生长因子表达失衡在关节炎的形成及发展过程中起重要作用,促血管生长因子表达升高是导致滑膜新生血管形成的主要原因。Objective The mechanism of growth factors in the formation of pannus is explored by detecting the expressions of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) and endostatin in synovium of collagen-induced arthritis (CIA) rats at different stages and determination of the micro-vessel density (MVD) by CD34. Methods After the model of CIA was established, the articular volume was measured and the synovium was examined with regular HE stain and the inflammation and pathological changes were evaluated. In addition, the VEGF, bFGF, endostatin and CD34 expressions in synovium were examined by Immunohistochemical staining. Results The expressions of VEGF and bFGF in CIA rats' synovium were evidently higher than that of the control group (P〈0.01), and the correlation was found between the expression of VEGF and bFGF (P〈0.01). The difference in the expressions of endostatin was not found between the experimental and control groups (P〉0.05). Conclusion Imbalance of the production of growth factors may play an important role in the development of arthritis. The increased expression of vascular growth factors may be the major cause of synovial pannus formation.
