摘　　要：目的探讨类风湿关节炎（RA）发病机制中促凋亡基因PDCD5的表达与作用。方法采用实时定量聚合酶链反应（PCR）和Westem blot方法来检测RA滑膜组织中PDCD5在mRNA和蛋白水平的表达，免疫组织化学检测PDCD5在滑膜内的表达特征，用骨关节炎（OA）患者滑膜标本作对照。结果RA滑膜组织中PDCD5 mRNA的表达为1．3±0．9，与OA滑膜组织（4．3±2．7）相比低了约4倍（P〈0．05），二者之间的差异有统计学意义。Western blot检测结果表明与OA滑膜组织相比较，RA滑膜组织中PD—CD5蛋白水平的表达较低。免疫组织化学染色显示在RA滑膜组织与OA滑膜组织之间PDCD5蛋白有不同的表达特征，RA滑膜组织阳性染色强度明显弱于OA滑膜组织，其增生滑膜表现为分散的PDCD5阳性染色．主要集中在衬下层而不是衬里层，衬下层主要分布为成纤维样滑膜细胞，而在OA滑膜中PDCD5阳性染色主要集中在衬里层。衬下层PDCD5表达的细胞较少。结论在RA滑膜细胞凋亡过程中PDCD5表现为下调，至少部分地减少了RA滑膜组织凋亡。Objective To investigate the expression of PDCD5 （programmed cell death 5, PDCDS） gene in the pathogenesis of rheumatoid arthritis （RA）. Methods The expressions of PDCD5 were evaluated at mRNA and protein level in synovial tissue of rheumatoid arthritis by real-time PCR, Western blot and immunohistochemistry respectively. Results Quantitative PCR revealed that low levels of PDCD5 mRNA were detected in RA synovial tissue （1.3±0.9） compared with osteoarthritis （OA） synovial tissue （4.3±2.7）, which had approximately 4-times more PDCD5 mRNA than RA in the synovial tissue （P〈0.05）. lmmunoblotting demonstrated that relatively lower expression of PDCD5 protein in RA synovial tissue compared with OA. Al- though PDCD5 expression was detected in the whole tissue sample, the expression pattern of PDCD5 protein were different beteeen RA and OA tissue.Generally, positive staining in OA synovium was evidently more intense than the RA synovial tissue.However, RA hyperplastic synovium showed a scattered pattern of PDCD5 expression, predominantly in the sublining rather than the intimal lining layer. In the contrast, in OA synovium, positive staining was predominantly seen in the lining layer, while PDCDS-expressing cells were less pronounced in the sublining. Conclusion The expression of PDCD5 could be down-regulated in the process of synovium apoptosis of RA, which had reduces the apoptosis of synovial tissue.
